Interaktionen der Hedgehog und Ras Signalwege beim Rhabdomyosarkom
Das Rhabdomyosarkom (RMS) ist das häufigste Weichteilsarkom bei Kindern. Wir konnten zeigen, dass der Hedgehog (Hh) Signalweg in Subgruppen dieser Tumoren sowohl beim Menschen als auch in der Maus aktiviert ist. Oft weisen RMS auch onkogene Ras Mutationen auf, die alle 3 Ras Gene betreffen können. Da Ras die Aktivität des Hh Signalwegs beeinflussen kann, untersuchen wir, ob onkogene Ras Mutationen das Wachstum von Hh-assoziierten RMS beeinflussen. Hierzu verwenden wir Zelllinien und Mausmodelle.

Die Rolle von Wnt5a beim Rhabdomyosarkom
Das Rhabdomyosarkom (RMS) entsteht aus undifferenzierten Muskelvorläuferzellen und ist das häufigste Weichteilsarkom bei Kindern. Obwohl der Wnt Signalweg eine wichtige Rolle bei der Entwicklung des Skelettmuskels spielt, gibt es nur wenige Informationen über seine Funktion beim RMS. Wir untersuchen die Rolle von Wnt5a, welches eine wichtige Komponente des ß-Catenin-unabhängigen Wnt-Signalwegs ist, in RMS Zelllinien und Mausmodellen.

Die Rolle des Hedgehog Signalwegs in der adulten Hypophyse und bei der Entstehung von Hypophysenadenomen
Der Hedgehog (HH) Signalweg spielt eine wichtige Rolle bei der Entwicklung der Hypophyse, einer Hormondrüse mit übergeordneter Rolle bei der Regulation des gesamten neuroendokrinen Systems. Über die Funktion dieses Signalwegs in der adulten Hypophyse und bei der Entstehung von Hypophysenadenomen ist wenig bekannt. Anhand von Mausmodellen und humanen Gewebeproben untersuchen wir, welchen Einfluss der Signalweg auf die Entstehung Hormon-exprimierender Zellen, die Hormonregulation und die Entstehung von Adenomen hat, und ob Smoothened-Inhibitoren für die Behandlung von Hypophysentumoren eingesetzt werden können.

Adulte Stammzellen der Haut und deren Rolle bei der Entstehung von Hautkrebs
Adulte Stammzellen der Haut sind essentiell für die natürliche Regeneration der Haut und für die Wundheilung. Jedoch spielen sie auch eine wichtige Rolle bei der Entstehung von Hauttumoren. In diesem Projekt untersuchen wir anhand von Mausmodellen und humanen Hautproben eine von uns erstmals beschriebene stammzellartige Zellpopulation der Haut und deren Funktion in der normalen Homöostase sowie deren Rolle bei der Entstehung von Hautkrebs.
